如何确定流式分析中荧光补偿是正确的呢？

How to determine the correct compensation during your Flow Cytometry data analysis?
The principle:
the median of the positives population is equal to the median of the negatives population.
也就是说要确定你的阳性细胞群和阴性细胞群的中位数要一致。背后的机理请看我的前一篇文章，荧光补偿基础知识
具体参见下图,
细胞染了CD3 FITC, 和 PE isotype. 水平的黑线表示细胞群的中位数中心。
figure 1 显示了，各种可能的补偿情况，你能知道那一个图是正确补偿的吗？

只有 阳性细胞群中位数＝阴性细胞群中位数 才是正确的补偿（必须要是单染的细胞样品）
Figure 2.
under compensated: 补偿不足, 既没有将所有的溢漏荧光减去，只补偿了一部分。
Over compensated: 过渡补偿，既矫正过渡。
警告： 肉眼补偿是非常危险的，很容易造成上述2种情况，一定要计算细胞群的中位数。
